Power and limitations of electrophoretic separations in proteomics strategies.

چکیده
Proteomics can be defined as the large-scale analysis of proteins. Due to the complexity of biological systems, it is required to concatenate various separation techniques prior to mass spectrometry. These techniques, dealing with proteins or peptides, can rely on chromatography or electrophoresis. In this review, the electrophoretic techniques are under scrutiny. Their principles are recalled, and their applications for peptide and protein separations are presented and critically discussed. In addition, the features that are specific to gel electrophoresis and that interplay with mass spectrometry (i.e., protein detection after electrophoresis, and the process leading from a gel piece to a solution of peptides) are also discussed.

Variations on a theme: changes to electrophoretic separations that can make a difference.
Electrophoretic separations of proteins are widely used in proteomic analyses, and rely heavily on SDS electrophoresis. This mode of separation is almost exclusively used when a single dimension separation is performed, and generally represents the second dimension of two-dimensional separations. Electrophoretic separations for proteomics use robust, well-established protocols. However, many va...
متن کاملImprovement of Capillary Electrophoretic Enantioseparation of Fluoxetine by a Cationic Additive
One of the problems encountered in CE separations of basic compounds is the adsorption of analytes onto the negatively charged capillary wall which could lead to poor repeatability of migration time and peak area. Additionally, separation of enantiomers of chiral of basic drugs is commonly carried out in low pH buffer which contributes to strong ionic interaction of the cationic drug ions with ...

متن کاملApplication of MALDI-TOF-mass spectrometry to proteome analysis using stain-free gel electrophoresis.
The combination of MALDI-TOF-mass spectrometry with gel electrophoretic separation using protein visualization by staining procedures involving such as Coomassie Brilliant Blue has been established as a widely used approach in proteomics. Although this approach has been shown to present high detection sensitivity, drawbacks and limitations frequently arise from the significant background in the...

It seems that geopolitics is the most important factor in Iran’s international and local evolutions. Iranians, in the line with political history, sometimes were the great global power as an empire, and sometimes were the local great power. For them, to be the great power in region, there are some causes that need to in mind. So opportunities in the path of being a great regional power are divi...
